The forkhead box P3 (FOXP3) transcription factor controls the function of regulatory T (Treg) cells, which actively maintain immune peripheral tolerance. Accordingly, FOXP3 mutations cause immunodysregulation, polyendocrinopathy, enteropathy, X-linked (IPEX) syndrome, a fatal autoimmune syndrome. Although FOXP3 expression is not restricted to Treg cells in the human system, its role in effector T cells has not been fully addressed.
