Loss-of-function mutations in CARD14 are associated with a severe variant of atopic dermatitis

Dominant gain-of-function mutations in CARD14, encoding a known regulator of NF-κB, cause psoriasis and related disorders. Here, the authors show that dominant negative mutations in the same gene result in severe atopic dermatitis and decreased NF-κB signaling.
