Analysis of coding and non-coding transcriptome of peripheral B cells reveals an altered interferon response factor (IRF)-1 pathway in multiple sclerosis patients

Several evidences emphasize B-cell pathogenic roles in multiple sclerosis (MS).We performed transcriptome analyses on peripheral B cells from therapy-free patients and age/sex-matched controls. Down-regulation of two transcripts (interferon response factor 1 –IRF1, and C-X-C motif chemokine 10–CXCL10), belonging to the same pathway, was validated by RT-PCR in 26 patients and 21 controls. IRF1 and CXCL10 transcripts share potential seeding sequences for hsa-miR-424, that resulted up-regulated in MS patients. (Source: Journal of Neuroimmunology)

Long-term persistence of anti-rods and rings antibodies in patients with chronic hepatitis C after antiviral treatment

AbstractAnti-rods and rings (anti-RR) antibodies are related to hepatitis C virus (HCV) in patients treated with pegylated interferon (PEG-IFN) and ribavirin (RBV). Only RBV induces rods/rings structures in vitro; but in vivo, the antibody appearance is related to the combination of these drugs, because data about patients using just one of these drugs alone is missing. Some studies suggest disappearance of these antibodies over time. The aim of this study was to describe the occurrence of anti-RR in patients with chronic hepatitis C treatment-na ïve or previously PEG-IFN/RBV-experienced, evaluating the persistence of anti-RR antibodies long after PEG-IFN/RBV treatment. From 2016 to 2017, 70 HCV-infected patients were screened for anti-RR using indirect immunofluorescence. Demographic and...

β-eudesmol inhibits thymic stromal lymphopoietin through blockade of caspase-1/NF-κB signal cascade in allergic rhinitis murine model.

Authors: Moon PD, Han NR, Lee JS, Kim HY, Hong S, Kim HJ, Yoo MS, Kim HM, Jeong HJ Abstract Allergic rhinitis (AR) is a global health problem because of its steadily increasing incidence and prevalence that currently affects about 30% of people worldwide. β-eudesmol has various beneficial effects, including anti-cancer and anti-allergic activities. However, the effects of β-eudesmol on AR have not yet been clarified; thus, we investigated the effects of β-eudesmol in an ovalbumin-induced AR animal model using enzyme-linked immunosorbent assay, histamine assay, Western blotting, and hematoxylin and eosin staining methods. β-eudesmol reduced the nasal rubs score and levels of histamine and immunoglobulin E in serum of AR mouse. In addition, the levels of thymic stromal lymphopoie...

Comparing Allergist And Cardiologist Considerations For The Optimal Management Of Thienopyridines Hypersensitivity.

Authors: Calogiuri G, Parlangeli C, Mirizzi AM, Macchia L, Foti C, Savage MP Abstract The thienopyridine family includes ticlopidine, clopidogrel and prasugrel which are antiplatelet drugs largely used, mainly associated to aspirin, for treatment of acute coronary syndromes and after percutaneous coronary interventions, to avoid thrombosis. In some patients, thienpyridines may cause hypersensitivity reactions which jeopardize the optimal therapeutic and preventive approach to vascular diseases. The management of thienopyridine hypersensitivity is best done as an interdisciplinary collaboration between the allergist and cardiologist. For the cardiologist, the important issues are the necessity of continuing therapy, the desired duration of therapy based on the clinical indication of...

PCAF fine-tunes hepatic metabolic syndrome, inflammatory disease, and cancer.

Authors: Wang T, Yao W, Shao Y, Zheng R, Huang F Abstract The P300/CBP-associating factor (PCAF), a histone acetyltransferase, is involved in metabolic and pathogenic diseases, particularly of the liver. The effects of PCAF on fine-tuning liver diseases are extremely complex and vary according to different pathological conditions. This enzyme has dichotomous functions, depending on differently modified sites, which regulate the activities of various enzymes, metabolic functions, and gene expression. Here, we summarize the most recent findings on the functions and targets of PCAF in various metabolic and immunological processes in the liver and review these new discoveries and models of PCAF biology in three areas: hepatic metabolic syndrome, inflammatory disease, and cancer. A comparison of CD105 and CD31 expression in tumor vessels of hepatocellular carcinoma by tissue microarray and flow cytometry.

In this study, we investigated the endothelial markers CD105 and CD31 in the tumor vessels from 90 patients with hepatocellular carcinoma (HCC) by tissue microarray, in addition to their expression in TECs isolated from fresh tissues resected from 11 patients with HCC by flow cytometry and confocal microscopy. The results revealed that among 90 cases of TMA, all tumor vessels were CD31 positive whereas 39 cases (43.3%) had little or no CD105 expression in tumors and their vessels but not peritumoral tissue spots, and that among these 39, 29 cases (74.4%) were poor-differentiated HCC. These findings were further verified by flow cytometry and confocal analysis of TECs isolated from HCC. Overall, the results suggested that CD105 may not be expressed in TECs derived from poor-differentiated H...

Comparison of sublingual immunotherapy and oral immunotherapy in peanut allergy

Abstract

The prevalence of food allergy has been increasing over the past few decades at an alarming rate with peanut allergy affecting about 2 % of children. Both oral immunotherapy (OIT) and sublingual immunotherapy (SLIT) have shown promise as a treatment option for peanut allergy. Immunotherapy induces desensitization and reduces the risk of reaction during accidental ingestion and may also enable those who are successfully desensitized to include the food allergen in their diet. OIT has been very well studied and has been found to be more efficacious than SLIT with an acceptable safety profile. However, SLIT is associated with fewer side effects. Studies indicate that a combination of SLIT and OIT may together induce a significant increase in challenge thresholds with fewer adverse events. More head-to-head clinical trials that directly compare OIT and SLIT as well as SLIT and OIT combination studies are warranted.

Verlängerung in Hessen erreicht!

The structure of situation models as revealed by anaphor resolution

Publication date: Available online 14 September 2018Source: Language SciencesAuthor(s): Sashank Varma, Amanda JanssenAbstractThis paper investigates the structure of situation models as revealed by the process of anaphor resolution. The situation model for a text combines the meanings of its individual sentences with inferences generated from prior knowledge into an overall understanding of the text. We conceptualize situation models as implemented by collections of traces in episodic long-term memory that are retrieved based on their similarity to cues in working memory. Traces encode both contextual (i.e., spatial and temporal) and semantic information, providing structure to situation models. Four predictions were derived regarding the retrieval of antecedent traces during anaphor resol...

Is there a pluralia tantum subcategory of nominal gerunds? Developing Gaeta's notion of morphological differentiation

Publication date: Available online 14 September 2018Source: Language SciencesAuthor(s): J. Lachlan MackenzieAbstractInspired by Gaeta's study of morphological distinctiveness as the basis for category formation, this article considers the possibility that by virtue of their invariable suffix -ings English pluralia tantum nominal gerunds (PTNGs) such as surroundings can be regarded as a subcategory of nominal gerunds. It is shown that the set of PTNGs is semantically very disparate with regard to the verb that underwent nominalization in the course of their formation. Differences are observed in a very large corpus with regard to the quantifiers and other items that co-occur with PTNGs, namely quantifiers regularly associated with plural count (e.g. many) or with mass nouns (e.g. much), but...

Social and configural effects on the cognitive dynamics of perspective-taking

Publication date: February 2019Source: Journal of Memory and Language, Volume 104Author(s): Alexia Galati, Rick Dale, Nicholas D. DuranAbstractHow do environmental cues and social perspectives influence perspective selection? Listeners responded to instructions (e.g., "Give me the folder on the right") from a simulated partner, selecting from two objects consistently aligned with themselves (ego-aligned; Experiment 1a) or the speaker (other-aligned; Experiment1b). In Experiment 2, listeners selected from triangular 3-object configurations whose orientation varied (ego-, other-, or neither-aligned). When the configural cue was other-aligned (consistently or inconsistently: Experiments 1b and 2), listeners were more likely to be other-centric. Other-centric responders stabilized their st...

A biotin receptor-targeted silicon(IV) phthalocyanine for in vivo tumor imaging and photodynamic therapy

Publication date: Available online 15 September 2018Source: Journal of Photochemistry and Photobiology B: BiologyAuthor(s): Ke Li, Wenyi Dong, Qingzhu Liu, Gaochao Lv, Minhao Xie, Xinchen Sun, Ling Qiu, Jianguo LinAbstractPhthalocyanines (Pcs) are a kind of potential photosensitizers for fluorescence imaging and photodynamic therapy (PDT). However, the clinical application of Pcs is suffered from their poor solubility, high aggregation tendency and low tumor-specificity. To address these problems, two biotin moieties were linked to the axial positions of silicon(IV) phthalocyanine (SiPc) through hydrophilic polyethylene glycol (PEG) linkers to synthesize a new water-soluble and tumor-targeting photosensitizer (compound 1). The introduction of PEG linkers on SiPc markedly reduced the aggreg...

On the neural basis of word reading: A meta-analysis of fMRI evidence using activation likelihood estimation

Publication date: February 2019Source: Journal of Neurolinguistics, Volume 49Author(s): K.A. Murphy, J. Jogia, J.B. TalcottAbstractThe neural networks that support normal single word reading have been studied extensively with functional magnetic resonance imaging (fMRI) and been heavily meta analysed; yet two caveats persist. Firstly, inclusion criteria are generally broad, allowing for a range of studies with inconsistent methodological practices to become a major source of variance in the data. Secondly, the sophistication of meta analytic techniques are ever improving, calling for a continuation of quantitative summaries for more informed interpretations of the literature. We conducted a novel meta-analysis using the updated activation likelihood estimation (ALE) approach on fMRI data f...

Parent-rating vs performance-based working memory measures: Association with spoken language measures in school-age children.

Authors: Magimairaj BM Abstract Working memory (WM) assessment is often part of cognitive-linguistic test batteries. The author examined the relationship between parent rating of WM functioning and children's performance-based WM, and the relationship of each with receptive and expressive spoken language measures. Study participants were eighty-three 7- to 11-year-old children. The sample represented a broad range of cognitive abilities. No participating child had frank neurological or intellectual disabilities, autism, fluency disorder, or hearing loss. Parents completed a standardized executive function rating scale that included a WM subscale. Children completed a standardized WM task and an experimental WM task. Children also completed six standardized language measures. Result...

A game of give and take: category change on the border between adverbial verbal gerunds and augmented absolutes in English

This article discusses one of several reasons for the loss of augmentation in English absolute constructions (ACs): the emergence of the verbal gerund (VG) in -ing. The VG was from the start strongly associated with prepositional contexts (de Smet, 2008) and has been shown to have a history connected to the AC (Fanego, 2004). More specifically, the strong association between VGs and prepositions probably caused a reinterpretation of most prepositionally augmented ACs as VGs, resulting in a functional split (Fonteyn & van de Pol, 2016). Second, this paper illustrates how the what with-construction underwent a very similar development, albeit in the opposite direction. Indeed, it appears that the grammaticalization of with into an AC-marker (van de Pol & Hoffmann, 2016), and the resulting st...

Facial Palsy: Diagnostic and Therapeutic Management

Facial expression is an evolutionary adaption facilitating successful social interaction. Muscles of facial expression are pivotal to nonverbal communication, articulation, and corneal protection. Facial palsy results in functional, communicative, and social impairment with profound negative impact on quality of life and emotional well-being. Congenital absence or acute facial nerve insult results in flaccid paralysis. Ultimate recovery following facial nerve insult lies on a spectrum from persistence of dense paralysis to return of normal function; in between zonal permutations of varying degrees of static and kinetic hypoactivity and hyperactivity and synkinesis exist.

Whether we like it or not, judgments based on facial appearance play a powerful role in how we treat others and how we ourselves are treated. What is striking is that the judgment during the first impression is made within 1/10th of one second.1

Describing the mandible in patients with craniofacial microsomia based on principal component analysis and thin plate spline video analysis

Craniofacial microsomia (CFM) is most often described as a unilateral malformation of derivatives of the first and second branchial arches. The mandible has been classified using several classification systems. However, all are based on two-dimensional imaging. The aim of this study was to mathematically describe the deformed mandible based on principal component analysis (PCA) in a three-dimensional way. This may aid in defining the flaws in existing surgical corrections of the mandible through the identification of the differences in shape compared with a normal mandible in a holistic view with the help of videos.

Zinc supplementation plays a crucial role in T helper 9 differentiation in allogeneic immune reactions and non-activated T cells

Publication date: Available online 6 February 2018Source: Journal of Trace Elements in Medicine and BiologyAuthor(s): Martina Maywald, Fudi Wang, Lothar RinkAbstractT helper (Th) 9 cells play a critical role in immune-mediated diseases, including allergic airway inflammation, autoimmune diseases, and cancer development. Thus, the promotion or suppression of Th9 cell differentiation, transcriptional control, and function is very important for a healthy immune system. Interestingly, T cell maturation, differentiation and function are highly dependent on the individuals' zinc status. This is especially seen in zinc deficient individuals as in the elderly population often suffering of autoimmunity and increased incidence of infections. In this regard, this study examines the impact of zinc su...

A pilot study on adjunctive use of parametric colour-coded digital subtraction angiography in endovascular interventions of haemodialysis access

Abstract

Background

Two-dimensional digital subtraction angiography (DSA) is the gold standard for angiographic evaluation of dysfunctional haemodialysis access. We aim to investigate the utility of parametric colour coded DSA in providing hemodynamic analysis during haemodialysis access interventions.

Methods

We retrospectively studied 20 patients who underwent access intervention and applied parametric colour-coding on selected DSA acquisitions before and after percutaneous transluminal angioplasty (PTA). The difference in time to peak (dTTP) contrast enhancement and time attenuation curve (TAC) of pre- and post-stenotic regions of interest (ROIs) were obtained and compared after treatment.

Results

Improvements were seen in mean percent of stenosis after PTA (p < 0.0001) for all cases. Median dTTP improved from 0.52 (IQR 0.26, 0.8) to 0.25 (IQR 0, 0.26) seconds (p = 0.001). Median 50% contrast washout time improved from 0.77 (IQR 0.39, 1.17) to 0.42 (IQR 0.23, 0.59) seconds (p = 0.031). Significant correlation was seen for dTTP vs. percent of stenosis (r = 0.723, p = 0.043) pre-PTA and for change in dTTP vs. percent change in stenosis post-PTA (r = 0.786, p = 0.021) for inflow lesions. Such correlation was however not seen in outflow lesions.

Conclusions

Adjunctive use of parametric colour-coded DSA may provide potentially useful hemodynamic information during vascular access interventions. Larger prospective studies are needed to validate our findings.

Generosity and Duration of Medicaid Expansion Waivers and Access to Care

Publication date: Available online 14 September 2018

Source: American Journal of Preventive Medicine

Author(s): Wafa W. Tarazi, Cathy J. Bradley, David W. Harless, Harry D. Bear, Lindsay M. Sabik

Introduction

Prior to expansion of Medicaid under the Affordable Care Act, some states obtained Section 1115 waivers from the federal government that allowed them to expand eligibility for Medicaid to adult populations that were not covered previously. Expansion waivers in these states differed in their generosity and year of implementation, creating variation in coverage availability and program longevity across states. This study examined the association between generosity and duration of Medicaid expansion waivers and access to preventive services.

Methods

The 2012 Medical Expenditure Panel Survey data were used to estimate adjusted logistic models in 2016, comparing outcomes among low-income non-elderly adults living in generous (Medicaid eligibility threshold ≥138% federal poverty level) and moderate (Medicaid eligibility threshold <138% federal poverty level) waiver states, relative to no-waiver states.

Results

Moderate and generous waivers were associated with statistically significant (p<0.001) increases in probabilities of having a usual source of care and a blood pressure check, relative to states without a waiver to expand. Low-income individuals living in states with longer waiver durations had better access to healthcare services than a similar group living in comparison states.

Conclusions

Not only is Medicaid waiver generosity associated with improving access to healthcare services, but the combination of generosity and longer duration of a waiver also intensifies the association. As states gain flexibility in designing their Medicaid programs, the healthcare benefits associated with both generosity and duration of waivers are important considerations for policy makers.

Reassessing Urban Health Interventions: Back to the Future with Google Street View Time Machine

Publication date: Available online 14 September 2018

Source: American Journal of Preventive Medicine

Author(s): Ronaldo L. Cândido, Madeleine Steinmetz-Wood, Patrick Morency, Yan Kestens

Introduction

Validity of research linking built environments to health relies on the availability and reliability of data used to measure exposures. As cities transform, it is important to track when and where urban changes occur, to provide detailed information for urban health intervention research. This paper presents an online observation method of the implementation of traffic-calmingfeatures using Google Street View Time Machine. The method is used to validate an existingadministrative database detailing the implementation of curb extensions and speed bumps.

Methods

Online observation of curb extensions and speed bumps was conducted for four boroughsin Montreal, Canada, in autumn 2016, and compared with administrative data documenting traffic-calming measures implemented between 2008 and 2014. All images available through the Time Machine function between 2007 and 2016 for 708 intervention sites were visualized online. Records in the administrative database were compared to real-world Google Street View observations and tested in terms of sensitivity, specificity, and positive predicted value.

Results

Google Street View Time Machine allowed the visualization of a median of seven different dates per street intersection and six dates per street segment. This made it possible to analyze built environment changes within 3,973 distinct time periods at 708 locations. Validation of the administrative data regarding presence of an intervention showed 99% (95% CI=97%, 99%) sensitivity, 58% (95% CI=51%, 64%) specificity, and 77% (95% CI=73%, 81%) positive predictive value.

Conclusions

Google Street View Time Machine allowed past (2007–2016) online documentation of microscale urban interventions—curb extensions and speed bumps. The proposed method offers a new way to document historic changes to the built environment, which will be useful for urban health intervention research.
